Development and validation of a UPLC method for 9 polyphenols determination in pear leaves

【Objective】A ultra performance liquid chromatography(UPLC)coupled with photodiode arraydetector(PDA)method for the detection of polyphenols in pear leaves was developed.【Methods】To de-termine the polyphenols of mature pear leaves,chromatographic separation conditions(mobile phase,concentration gradient and detection wavelength)of the UPLC method was optimized and validated.【Re-sults】Mobile phase A: 0.5% formic acid solution,mobile phase M: acetonitrile. 9 polyphenols were si-multaneously separated under the condition of 0-1 min,0-5% B;1-6 min,5%-17% B;6-10 min,17%-25% B;10-11 min,25%-100% B;11-14 min,100% B;14-15 min,100%-0 B;15-20 min,0 B;andthe chromatograms were acquired at 280 nm,325 nm and 350 nm. Liner regression equations of polyphe-nols were tested,of which correlation coefficients were more than 0.999 4. The recoveries ranged from83.39% to 94.31%. Relative standards deviations of precision were less than 4.77%.【Conclusion】Thismethod has good accuracy,high precision and suitable for simultaneous detection of 9 polyphenols inpear leaves.
