Extraction and analytical methods of carotenoids in fruit of yellow flesh peach

【Objective】In order to establish an efficient and speedy methods for extracting and analyzingcarotenoids with High-performance liquid chromatography(HPLC) in fruit of yellow flesh peach.【Meth-ods】We optimized the extraction and analytical methods of carotenoid from following several steps: ratioof material to liquor,extraction method and time,proportion and speed of mobile phase and quantity ofinjection. And also,we evaluated the determination method.【Results】The fruits flesh extracted with ace-tone,the ratio of material to liquor was 1∶6(g·m L-1),the samples were extracted by an ultrasonic methodfor 1 h with darkness;the samples were measured by YMC-C30 column,sample introduction was 20μL,solvents were Methyl Tertiary Butyl Ether and methanol(V/V=30∶70) with type of isocratic elution,the flow rate was 1.0 m L·min-1and temperature was 25 ℃,analyzing with UV at 450 nm. The extractionand determination methods we established could extract and detect carotenoids effectively and speedily,with the RSD for the accuracy 0.99%-2.25% and 95%-105% recovery rate. The extraction and determi-nation system has advantages of simple operation,high efficiency,good stabilization,high resolution,ac-curate result,shorter time and lower cost.【Conclusion】The methods established in this study were per-fectly viable for the extraction and determination of carotenoids in fruits of yellow flesh peach.
