【Objective】In order to provide a rationale for the ripening mechanism study of pear fruit,thepectate lyase gene of‘Korla fragrant pear'(Pyrus sinkiangensis Yu),named Ps PL,was isolated,the ex-pression profiles of Ps PL were monitored in different periods.【Methods】The full length c DNA sequence of Ps PL was isolated from the fruit tissue of‘Korla fragrant pear'with the method of RT-PCR and RACE.We predicted the function of Ps PL by bioinformatics,the expression characteristics of this gene in lategrowth period and shelf life period were analyzed using q RT-PCR(quantitative real-time PCR).【Re-sults】RACE result showed that the c DNA sequence of Ps PL is 1 245 bp in full length and it codes a pre-dicted protein of 414 amino acids(Gen Bank Accession No. KJ547669). Bioinformatics analysis showedthat Ps PL belong to the pectate lyase superfamily,the amino acid identity compared with other plant PL homologues is highly conserved and shows the highest similarity(97%)with the PL of Malus×domestica.The result of q RT-PCR showed that the expressions of Ps PL in different periods were very different,itwas very low at late growth periods and reached a high level at 150 days after full bloom.【Conclusion】Thepectate lyase gene,Ps PL,which was isolated from‘Korla fragrant pear',may play an important role dur-ing fruit softening in‘Korla fragrant pear'.
