Molecular cloning and expression of PyCHI and PyF3H in‘Pingguoli'during the pigmentation

【Objective】To clone the full-length sequences of the c DNA of Py CHI and Py F3 H from‘Ping-guoli',and to analyze the relationship between the expression and the accumulation of anthocyanin dur-ing the pigmentation of‘Pingguoli'.【Methods】With the‘Pingguoli'as material,the full-length c DNAsequences of Py CHI and Py F3 H were cloned by the homologous cloning technology. The content of antho-cyanin was determined and the expressions of Py CHI and Py F3 H were respectively analyzed by real-timefluorescent quantitative PCR technique during the pigmentation.【Results】Sequence analysis showed thatthe c DNA of Py CHI and Py F3 H were 702 bp and 1095 bp in length,respectively,encoded 233 and 364 amino acids. The similarity between these c DNA fragments and the genes of other Rosaceous plants ismore than 95%,the amino acid sequences of them had more than 85% similarity. Real-time fluorescent quantitative PCR indicated the expression levels of Py CHI and Py F3 H were markedly enhanced in thesunlit side,then the Py CHI decreased slightly. However,their expression levels were higher than that ofcontrol group during the pigmentation. Thevariation trends were similar with the content of anthocyanin.【Conclusion】Full- length c DNA of Py CHI and Py F3 H were cloned,and their expression levels wereclosely related to the synthesis of anthocyanin,and might play important roles during the pigmentation of‘Pingguoli'.
